Project Title: Antigen specific memory B cell responses in Bangladeshi adults after one or two-dose oral killed cholera vaccination and comparison with responses in patients with naturally acquired cholera.

Project Title: A Phase I Double-Blinded, Placebo-Controlled, Dose Escalating study to evaluate the safety and Immunogenicity of Double Mutant Heat-Labile Toxin LTR192G/L211A (dmLT) from ETEC by Vaccination in Adults residing in an endemic area.

Project Title:  Correlates of protection of COVID-19 immunology (COPCI) study.

Molecular Techniques:

● Isolation of Peripheral Blood Mononuclear Cells (PBMCs) and plasma from whole blood by using Ficoll-Isopaque density gradient centrifugation

● ELISPOT assay for antigen specific Antibody Secreting Cells (ASCs)

● Evaluation of Antibody in Lymphocyte Supernatant  (ALS) samples for detection of active cholera infection

● Measurment of antigen specific gut homing ASCs

● Cryopreservation and thawing of the PBMCs

● Memory B cells (MBC) culture

● ELISPOT assay for antigen specific MBC in PBMCs

● Counting of antigen specfic MBC and ASCs by CTL autoanalyzer

● ELISA for LPS Ogawa, Inaba and O139 specific IgA, IgG and IgM antibodies responses in vaccinees

● Vibriocidal antibody assay in plasma

● Immunophetyphing of the MBC and plasmablast cells by Fluorescence-activated Cell Sorting (FACS) assay and analysis of the data via FlowJo software

● Immunophenotyping of memory T cells containing SARS-CoV-2-specific T cells determined by flow cytometry assay
